چکیده

Profilin-1 (PFN-1) is an actin-binding protein that regulates actin polymerization, cell proliferation, apoptosis, angiogenesis, and carcinogenesis. Its dysregulation has been reported in diverse pathologic diseases; however, the role of PFN-1 psoriasis not yet elucidated. In this study, we demonstrate expression increased both skin serum patients with psoriasis. was markedly expressed epidermis psoriatic lesions its positively correlated severity. The human keratinocytes induced by IL-17A TNF-α stimulation, but only presence secreted out cell. addition, knockdown shRNA resulted altered psoriasis-associated inflammatory markers, HBD-2, S100A7, S100A9, Ki67, recombinant suppressed IL-17A-induced response keratinocytes. Interestingly, also IκBζ, important player immune Considering these facts, it suggested balance intracellularly or extracellularly for IκBζ regulation pathogenesis Our results showed acts as a negative regulator inflammation through suppression IκBζ. Furthermore, epidermal hyperplasia observed psoriasis-like reconstructed equivalents were reduced PFN-1, which indicates aberrant development due to response. conclusion, our findings can be used biomarker disease severity, allude considered possible target suppressing preventing onset alleviating symptoms.

IκBζ is a key driver in the development of psoriasis.

Psoriasis is a common immune-mediated, chronic, inflammatory skin disease characterized by hyperproliferation and abnormal differentiation of keratinocytes and infiltration of inflammatory cells. Although TNFα- and IL-17A-targeting drugs have recently proven to be highly effective, the molecular mechanism underlying the pathogenesis of psoriasis remains poorly understood. We found that expressi...

Unilateral Hypothalamus Inactivation Prevents PTZ Kindling Development through Hippocampal Orexin Receptor 1 Modulation

Introduction: Epilepsy is a neural disorder in which abnormal plastic changes during short and long term periods lead to increased excitability of brain tissue. Kindling is an animal model of epileptogenesis which results in changes of synaptic plasticity due to repetitive electrical or chemical sub-convulsive stimulations of the brain. Lateral hypothalamus, as the main niche of orexin neurons ...

Regulation of endothelial cell adhesion by profilin

BACKGROUND Although profilin is believed to be an essential regulator of the actin cytoskeleton in most cells, its precise role in mammalian cells remains unknown. We have used replication-incompetent adenovirus carrying the human profilin I cDNA as a means rapidly to increase the concentration of profilin in human aortic endothelial cells 12-31-fold above baseline--levels never before achieved...

Novel mutations support a role for Profilin 1 in the pathogenesis of ALS

Mutations in the gene encoding profilin 1 (PFN1) have recently been shown to cause amyotrophic lateral sclerosis (ALS), a fatal neurodegenerative disorder. We sequenced the PFN1 gene in a cohort of ALS patients (n = 485) and detected 2 novel variants (A20T and Q139L), as well as 4 cases with the previously identified E117G rare variant (∼ 1.2%). A case-control meta-analysis of all published E11...
